The application of PVP in pharmaceutical research and development
PVP has become a very commonly used pharmaceutical polymer material in pharmaceutical research and development, and its advantages have been more and more recognized, making it a mature multi-purpose pharmaceutical excipients. PVP is a water-soluble polymer catalyzed by N-vinyl-2-pyrinolanone monomer, which is easily hygroscopic and easily soluble in polar organic solvents such as water and ethanol. The molecular weight of medicinal povidone is between 10,000 and 700,000, generally expressed by the size of K, the larger the K, the higher the molecular weight. PVP has many excellent properties, and its application in pharmacy is more and more extensive, more and more applicable, the following is the main aspects of its application in the development and research of pharmaceutical preparations in China.

PVPK15 (8,000)
PVPK25 (30,000)
PVPK30 (50,000)
PVPK60 (400,000)
PVPK90 (1,000,000)

1. Adhesive for oral solid preparation
PVP is soluble in water and some commonly used organic solvents, making it suitable for use in many situations. First of all, in the drug sensitive to humidity and heat, the use of PVP ethanol solution as a binder can eliminate the influence of water and heat, and for hydrophobic drugs, its aqueous solution can be uniformly wetted, and the surface of drug particles can be hydrophilic, which is conducive to increasing the dissolution of drugs. In addition, PVP as an adhesive can be added with dry powder, wetting granulating with appropriate solvent or direct pressing of dry adhesive can improve the phenomenon of hydrophobic drug stripping. K30 and K25 are commonly used, and the amount of PVPK30 is generally 2%-5%.
In the use of effervescent agents, PVP is an ideal adhesive, because effervescent agents require strict control of water content, using PVP ethanol solution as a binder can solve this problem. The effervescent tablets made by this method have good obviousness, rapid dissolution and strong foaming. With about 5%-10% PVP solution as binder can also be used for fluidized bed spray drying granulation.
2, used in oral liquid preparations to thicken and viscose
PVP has a long history as a thickening agent in oral liquid preparations and has a wide range of applications, such as its emulsion and suspension PVPK-90 can be used as a good thickening stabilizer, and the polymer is adsorbated on the surface of a single colloidal ion to form a thin molecular layer to prevent its coalescence.
3, used as a solid dispersion carrier
To improve the absorption of insoluble drugs in vivo has always been a hot topic, to achieve this purpose, consider improving its solubility and dissolution rate, solid dispersion is the insoluble drugs in a molecule, colloidal, microcrystalline or amorphous form dispersed in a water-soluble material, can significantly improve the dissolution rate and more absorption. PVP is a common carrier of vegetation solid dispersion.
