2-cyanopyrazine is an important organic chemical raw material and pharmaceutical intermediate, which has been widely used in the fields of fragrance, medicine and so on. Cyanopyrazine is the basic raw material for the synthesis of pyrazinamide tuberculosis special medicine.
Synthesis method
In a 1-liter three-necked flask, 500 ml of toluene, 48 g (0.3 mol) 2-bromopyrazine, 11.8 g (0.36 mol, 1.2 eq) sodium cyanide, 5.73 g (30 mmol, 0.1 eq) cuprous iodide, 10 g potassium iodide (60 mmol, 0.2 eq), 26.4 g N, were successively added under nitrogen protection. N ‘-dimethylethylenediamine (0.3 mole, 1.0 equivalent) was stirred under nitrogen protection at 110℃ for 30 hours to complete the reaction, and then filtered, the filtrate was fractionated under reduced pressure to obtain a transparent liquid 2-cyanopyrazine with 70% yield and 99% purity (GC). The boiling point was 84-87 ℃(18-20mmHg).
