Crosslinked polyethylpyrrolidone (PVPP) is a kind of crosslinked polymer which is polymerized from vinylpyrrolidone monomer under certain conditions and is insoluble in water, strong acid, strong base and common organic solvents. General properties and solubility
Crosslinked polyvinylpyrrolidone PVPP, white or nearly white, with hygroscopicity and easy to flow powder, odorless or slightly odorless, insoluble in water, alkali, acid and common organic solvents, with strong expansion performance and complex ability with many kinds of substances. The water absorption rate increases with the increase of relative humidity, and the formation of gel inhibits the penetration of water into the tablets, and the disintegration rate of the tablets slows
down. PVPP, in contrast to some other disintegrating agents, is insoluble and has no tendency to form gels at all.
Complexation and solubilization
PVPP and a variety of medicines and other substances can form complexes, the formation process is reversible, in the alkaline medium is not formed. It is more likely to form complexes with those aromatic compounds containing phenyl and/or carboxylic groups, and therefore, in the pharmaceutical industry, it can improve the solubility and bioavailability of medicines, improve the taste of perpyrexine, adsorb polyphenols and tannins from tinctures and herbs, and has the effect of combating diarrhea, gastritis, ulcers, esophageal hiatus cancer, etc.
Swelling and disintegration
PVPP has high capillary activity, so it can quickly absorb water into the tablet. Because the internal pressure (swelling pressure) exceeds the strength of the tablet,the tablet dissolves in an instant. And because there are folded molecular chains between the crosslinks, when water or aqueous solutions penetrate, they are stretched by the sudden impact and forced to separate immediately. The expansion results in an increase in net volume (approximately twofold in 5 minutes),resulting in the immediate disintegration of tablets formulated with PVPP tablet disintegrators.
Compressibility and adhesion
PVPP has excellent fluidity and plastic deformation, so they have excellent bonding properties. This property gives good compressibility in spray, dry, and wet granulation. When PVPP tablet disintegrators are added to tablets, compared with commonly used starch, cellulose preparations and pectin compounds, the hardness and wear resistance of tablets are not affected, and the cap length of tablets is not reduced. PVPP can also be used effectively as a tablet disintegrator in direct compression tablets without the need for lubricants or adhesives.
Capillary activity and hydration capacity
Studies have shown that PVPPS as tablet disintegrators have high capillary activity and hydration capacity due to their large specific surface area (average 1.25 m/g) and significant water absorption (high and rapid water absorption, up to more than 60%).
Good compatibilities
PVPP and a variety of organic and inorganic pharmaceutical excipients and active ingredients have very good compatibability, and solid drugs containing PVPP tablet disintegrators can be easily produced using common pill manufacturing techniques, including direct tablet pressing, spray, drying and wet granulation.
